Company overview

Royalty Pharma plc provides capital to universities, research institutions and drug companies by buying their rights to future drug-sales payments or funding development in exchange for new rights. Most of its portfolio consists of royalty rights bought from existing holders. Drug marketers’ sales generate the payments: Royalty Pharma generally receives a contractual share of sales, not profits, and uses cash from existing royalties to buy new investments.

The portfolio covers marketed medicines and development-stage candidates. Vertex’s cystic fibrosis medicines, including Trikafta/Kaftrio and Alyftrek, are its largest income source, and a handful of leading product franchises generate most royalty receipts. Royalty Pharma is disputing Vertex’s payments on Alyftrek, which it believes fall short of its contractual entitlement.

The companies developing and marketing the medicines generally control trials, approvals, manufacturing and sales; Royalty Pharma generally cannot direct their work. Its income therefore depends on their ability to bring medicines to market and sustain sales. Development-stage investments may never reach approval or generate royalties.

To remain outside U.S. investment-company regulation, Royalty Pharma and its subsidiaries must maintain a qualifying asset mix under the exclusions they rely on. This limits which assets they can hold or acquire.

Royalty Pharma depends on its portfolio’s drug sales and development outcomes, so also watch Roche’s Evrysdi and privately held Servier’s Voranigo, whose marketers are missing from this list.

  • Vertex sells cystic fibrosis medicines and pays Royalty Pharma a share of sales, making it the source of Royalty Pharma’s largest franchise. Watch Alyftrek’s royalty dispute: Vertex says the rate is 4%, while Royalty Pharma seeks approximately 8%.

  • Ligand competes with Royalty Pharma to fund drug developers and buy rights to a share of medicine sales. Its royalty purchases and development financing show where a rival is putting capital to build future royalty income.

  • GSK’s sales of the respiratory medicine Trelegy feed Royalty Pharma’s sales-based payments, contributing $332 million of receipts in 2025. Trelegy sales are the GSK revenue line to watch for this exposure.

  • Biogen’s Tysabri and Spinraza sales generate royalties for Royalty Pharma, which also funds Biogen’s litifilimab development. Watch those two medicines’ sales and litifilimab’s progress: Royalty Pharma provided $200 million of development funding in 2025.

  • Johnson & Johnson’s sales of Tremfya and jointly marketed Imbruvica support Royalty Pharma’s royalty income; together they generated $348 million of receipts in 2025. Tremfya sales forecasts matter beyond cash receipts: higher forecasts helped lift Royalty Pharma’s expected royalty cash flows in 2025.

  • Revolution Medicines receives up to $2 billion from Royalty Pharma through rights to future daraxonrasib sales and a secured loan, tying Royalty Pharma to this cancer drug’s success. FDA approval is a key event to watch because it requires Revolution Medicines to draw the first loan tranche.

  • Cytokinetics receives launch financing from Royalty Pharma and has sold it shares of revenue from MYQORZO, omecamtiv mecarbil and ulacamten. Watch MYQORZO’s U.S. sales to its focused cardiology customer base as Cytokinetics turns its launch into revenue.

  • Pfizer co-markets Xtandi, so sales of this prostate cancer medicine feed Royalty Pharma’s royalties and generated $197 million of receipts in 2025. Higher Xtandi forecasts also increased Royalty Pharma’s expected royalty cash flows that year.

  • Novartis sells Promacta/Revolade under an agreement with Royalty Pharma, which collects a share of those medicine sales. Promacta generated $142 million of Royalty Pharma’s receipts in 2025, making its sales the relevant Novartis revenue line to follow.

  • AbbVie co-markets Imbruvica, whose sales generated $170 million of Royalty Pharma’s receipts in 2025. Watch Imbruvica sales amid competition from other blood cancer treatments, rather than treating AbbVie’s overall results as a proxy for these royalties.

  • Amgen’s Imdelltra sales and olpasiran development matter to Royalty Pharma, which owns royalty interests in both medicines. Watch Imdelltra sales outside China for current payments and olpasiran’s development for future royalty potential.

  • Ionis sold Royalty Pharma a minority share of future Spinraza and pelacarsen royalties for $500 million upfront, so both participate in those medicines’ success. Watch pelacarsen approval and sales milestones: they determine additional payments to Ionis, with some subject to deadlines.

  • Exelixis relies mainly on cabozantinib revenue and pays Royalty Pharma 3% of its partners’ net sales of cabozantinib medicines. Watch those sales outside the U.S.: Ipsen sells outside the U.S. and Japan, while Takeda sells in Japan.

  • Syndax received $350 million from Royalty Pharma in return for 13.8% of U.S. Niktimvo net sales, subject to adjustments, until payments reach $822.5 million. Its U.S. Niktimvo sales, co-commercialized with Incyte, show how quickly Royalty Pharma receives payments toward that cap.

  • Ascendis received $300 million from Royalty Pharma in exchange for capped shares of U.S. SKYTROFA and YORVIPATH sales. Watch U.S. net revenue from each medicine: Royalty Pharma receives 9.15% from SKYTROFA and 3% from YORVIPATH under the separate agreements.

  • Ultragenyx sells Crysvita, whose sales and forecasts affect both Royalty Pharma’s royalty receipts and the value of its royalty assets. Lower Crysvita forecasts contributed to Royalty Pharma’s expense for reduced expected cash flows in 2024.

  • Takeda’s Entyvio sales and Japanese cabozantinib sales support Royalty Pharma’s royalties. Watch Entyvio’s sales for ulcerative colitis and Crohn’s disease, and Japanese cabozantinib sales, on which Exelixis pays Royalty Pharma a share.

  • Gilead sells Trodelvy, and those medicine sales generate royalty receipts for Royalty Pharma. Trodelvy sales are the Gilead revenue line to watch for this connection.
