Ligand Pharmaceuticals buys rights to a share of medicine sales, funds drug development in exchange for those rights, and acquires businesses or assets that already carry them. It also licenses its technologies to drug companies. Partners develop, manufacture and sell the medicines, then pay Ligand under their agreements. These royalties provide most of its revenue, with important sources including Amgen’s Kyprolis, Recordati’s Qarziba and Travere’s Filspari.
Ligand also sells Captisol, a material that helps drugs dissolve and remain stable, to partners that use it in their medicines. Hovione manufactures Captisol for Ligand, making that supply relationship important to this business. Technology licensing and development-stage agreements also bring license fees and payments when specified milestones are reached.
The royalty business depends on partners advancing drug programs, securing regulatory approval and selling approved medicines. Drug candidates cannot be marketed without approval from the FDA or the relevant overseas regulator. Under most Captisol licenses, royalties fall or stop when the relevant patents expire. Ligand also has to keep its asset mix within the limits of exemptions it uses to avoid registration under the U.S. Investment Company Act, which constrains how it can build its investment portfolio.
The risk section is the longest, dullest part of the filing. This keeps only what can really hurt you: where it hits, how hard, when it plays out.
Every company claims an edge. Each one is checked against hard numbers like margins and returns: which hold, and which line gives way first.
Watch Recordati even though it is absent from this list: its Qarziba sales generated $33.7 million of Ligand’s royalty receipts in 2025.
Travere sells the Ligand-licensed kidney medicine Filspari and pays Ligand 9% of worldwide sales, so Filspari sales directly feed Ligand’s income. Those payments generated $32 million of royalty receipts in 2025.
Amgen’s Kyprolis sales pay Ligand twice: Amgen buys Captisol for the medicine and pays royalties under its license. Watch Kyprolis sales, which generated $35.5 million of Ligand royalty receipts in 2025, separate from Captisol material revenue.
Merck’s sales of the vaccines Capvaxive and Vaxneuvance and the respiratory medicine Ohtuvayre feed Ligand’s royalty income. These product sales, rather than Merck’s overall revenue, are the lines to watch for Ligand’s receipts.
Royalty Pharma plc competes with Ligand to fund biopharmaceutical rights holders in exchange for a share of drug sales, including medicines still in development. Its purchases of drug royalty rights show where another buyer is deploying capital in Ligand’s market.
Jazz sells Rylaze using rights obtained from Ligand’s Pfenex business, so sales of this leukemia and lymphoma medicine contribute to Ligand’s royalty income. Rylaze generated approximately $13.3 million of Ligand royalty receipts in 2025.
Pelthos pays Ligand 13% of worldwide Zelsuvmi sales for its licensed NITRICIL technology, while Ligand also holds a substantial equity interest in Pelthos. Watch sales of this molluscum contagiosum treatment to wholesalers, which supply pharmacies.
Gilead uses Ligand’s Captisol in its COVID-19 medicine Veklury, so demand for Veklury matters to Ligand’s material-supply revenue.
Palvella’s Qtorin rapamycin programs could pay Ligand milestones and 8%–9.8% sales royalties if development succeeds. Palvella had no approved products or product revenue at December 31, 2025, so watch progress toward approval and its ability to meet milestone payments before sales begin.
Agenus owes Ligand a contractual share of future worldwide BOT/BAL net sales—a 2.625% synthetic royalty subject to adjustments—and shares specified milestone and royalty receipts from other partnered programs. Watch BOT/BAL’s cancer-treatment progress; Agenus began earning treatment revenue through authorized access programs in 2025, but those programs are not marketing approval.
LeonaBio licenses lasofoxifene rights from Ligand, giving Ligand potential milestones and 6%–10% royalties on future net sales if the breast-cancer medicine reaches commercialization. Watch its registrational Phase 3 trial in metastatic breast cancer with ESR1 mutations; LeonaBio had no approved drugs or drug-product revenue through December 31, 2025.
Orchestra BioMed received Ligand financing in exchange for tiered payments on Orchestra’s revenues from AVIM Therapy and Virtue SAB—not on the products’ total sales. Watch revenues reaching Orchestra from these hypertension and artery-disease programs, since those are the basis for Ligand’s payments.
Ligand Pharmaceuticals Inc. earningsThis company
Ligand earns mainly from royalties on partners’ drug sales, and this report will show whether royalties and Captisol sales support the business rather than contract income alone.
Read royalty revenue and Captisol sales separately from contract income. If the first two hold up or grow, partner-drug sales and material orders are supporting revenue. If they weaken while contract income props up the total, check whether milestones or transaction-related income that may not recur are masking the change. The question is whether Ligand’s ongoing revenue sources are keeping pace with reported revenue.
Travere Therapeutics, Inc. earningsTVTX
Travere’s Filspari sales feed directly into Ligand’s royalty income, so its report will show whether demand for the drug is growing or weakening.
Look at Travere’s reported Filspari sales, then compare them with the related royalty income Ligand reports. Rising Filspari sales expand the base for Ligand’s payments under the agreement; flat or falling sales put that income under pressure. Filspari generated $32 million in royalty receipts for Ligand in 2025. The question is whether the drug can remain a meaningful source of Ligand income.
Amgen Inc earningsAMGN
Amgen’s Kyprolis sales generate royalties for Ligand, and its report will show the demand behind one of Ligand’s largest royalty streams.
Watch Amgen’s reported Kyprolis sales, not Amgen’s total revenue as a proxy for Ligand’s results. Higher Kyprolis sales would support Ligand’s related royalty income; lower sales would weaken that source. Kyprolis generated $35.5 million in royalty receipts for Ligand in 2025. The question is whether this important product income is holding up.
Royalty Pharma plc earningsRPRX
Royalty Pharma also funds drug royalty rights, and its report will show how actively it is buying assets that Ligand may also pursue.
Look at the acquisitions and capital deployment Royalty Pharma reports. If it keeps buying drug royalty rights actively, Ligand may face stronger competition for suitable assets and tougher deal terms. If purchases slow, that pressure may ease, but it would not prove asset prices have fallen. The question is whether competition for Ligand’s acquisitions is intensifying.
