Company overview

Syndax Pharmaceuticals has two FDA-approved medicines on the U.S. market. Revuforj treats eligible patients whose acute leukemia has returned or resisted treatment and carries specified KMT2A or NPM1 genetic changes; the NPM1 indication also requires that patients have no satisfactory alternative treatment. Niktimvo treats eligible patients with chronic graft-versus-host disease after multiple prior systemic treatments have failed.

Most revenue comes from Revuforj, for which Syndax manages sales, marketing and distribution. Niktimvo provides a second substantial source: Syndax co-commercializes it with Incyte in the United States and shares the resulting profits and losses equally. Incyte leads commercialization globally and holds exclusive rights outside the United States, where Syndax is eligible for sales royalties and milestone payments. Syndax also sold Royalty Pharma a right to a portion of U.S. Niktimvo sales in exchange for upfront funding, with total payments capped.

The business depends on government and private insurers covering these medicines, Incyte's work on Niktimvo, and outside suppliers maintaining drug supply. Syndax has no manufacturing facilities of its own and relies on contract manufacturers and Incyte. It may promote the medicines only for approved uses and consistently with their FDA labeling.

Syndax’s results hinge on Revuforj adoption and Niktimvo commercialization with Incyte, while Niktimvo also depends on its underlying license from UCB.

  • Kura Oncology’s KOMZIFTI competes with Syndax’s Revuforj for adults with relapsed or refractory AML carrying a susceptible NPM1 mutation who have no satisfactory alternative treatment options; both are oral menin inhibitors. Kura’s U.S. KOMZIFTI sales are a concrete measure to watch as this competing treatment gains use.

  • Syndax depends on Incyte to lead global Niktimvo commercialization and supply product, and the two share U.S. commercialization profits and losses equally. Watch Niktimvo sales alongside production and shared selling costs: Syndax records its half of the resulting profit as collaboration revenue, not half of product sales.

  • Syndax pays AbbVie milestones and sales royalties for the right to develop and sell its menin-inhibitor assets, so those obligations accompany the business’s progress. AbbVie also markets Venclexta in AML, placing it among the treatments competing with Syndax’s Revuforj.

  • Johnson & Johnson’s Janssen menin-inhibitor program could challenge Syndax’s Revuforj, making its development progress relevant to Revuforj’s competitive position. This is a competing drug program to watch, rather than a shared revenue stream.

  • Rigel Pharmaceuticals’ REZLIDHIA and Syndax’s Revuforj both treat relapsed or refractory AML, but their approved mutation-defined patient groups differ. REZLIDHIA serves adults with a susceptible IDH1 mutation, while Revuforj’s AML indication covers patients aged one year or older with a susceptible NPM1 mutation and no satisfactory alternative treatment options, so REZLIDHIA sales are not a like-for-like measure of competition.

  • SELLAS Life Sciences develops AML treatments that could compete with Syndax’s marketed Revuforj, so AML trial outcomes matter to both businesses. Watch SELLAS’s cancer-drug development results for signs of future competition, rather than treating it as an existing commercial rival.
