Trevi Therapeutics is a clinical-stage drug developer focused entirely on Haduvio, an experimental extended-release nalbuphine tablet. It is developing the drug for chronic cough associated with idiopathic pulmonary fibrosis and other interstitial lung diseases, and for chronic cough that has not responded to other treatments. Its most advanced program targets cough associated with idiopathic pulmonary fibrosis. Trevi has no approved products and has generated no revenue.
Trevi retains worldwide commercial rights to Haduvio. If approved, it plans to sell the drug in the United States through its own specialist sales force and use third-party arrangements elsewhere. Sales would depend on physicians prescribing it and health insurers and other payors providing coverage and reimbursement. Trevi could promote it only for approved uses, not for cough generally.
The business depends on completing Haduvio’s clinical development, obtaining regulatory approval and raising additional capital. Trevi outsources manufacturing and relies on Par Health as its sole supplier of the active ingredient, with no qualified alternative or agreement guaranteeing supply quantities or prices.
The risk section is the longest, dullest part of the filing. This keeps only what can really hurt you: where it hits, how hard, when it plays out.
Every company claims an edge. Each one is checked against hard numbers like margins and returns: which hold, and which line gives way first.
Trevi’s prospects hinge on Haduvio’s clinical results and regulatory approval, while privately held Nocion Therapeutics is developing a competing treatment for refractory chronic cough.
GSK’s camlipixant competes for the refractory-chronic-cough patients Trevi aims to treat with Haduvio. GSK sells medicines and vaccines, but its camlipixant clinical results are the news most directly relevant to Haduvio’s competitive position.
Merck’s gefapixant already competes with Trevi’s intended use for Haduvio: it is approved for refractory or unexplained chronic cough in Japan, the UK, Switzerland and the EU. Merck sells prescription medicines to buyers including wholesalers and hospitals, but this cough treatment’s competitive reach excludes a planned US launch: Merck withdrew its FDA application and said it did not plan to refile.
Avalyn Pharma’s treatments for lung fibrosis could, if approved, reduce demand for Haduvio to treat the associated chronic cough; like Trevi, Avalyn is still developing its medicines and has no approved products or product-sales revenue. Watch AP01’s Phase 2b development for progressive pulmonary fibrosis and AP02’s Phase 2 development for idiopathic pulmonary fibrosis.
GSK plc earningsGSK
If GSK’s report updates camlipixant’s clinical progress, it will show how far GSK’s competing program for refractory chronic cough has advanced against Trevi’s Haduvio.
Look for any disclosed progress or setback in camlipixant’s clinical development. Continued progress would make GSK a more immediate competitor for the refractory-cough patients Trevi aims to treat, increasing the pressure on Haduvio to win patients; a delay or setback would ease that pressure for now, but would not show that Haduvio works or will be approved.
Avalyn Pharma Inc. earningsAVLN
If Avalyn Pharma’s report updates its AP01 and AP02 lung-fibrosis programs, it will show whether treatments that could affect future demand for Trevi’s cough drug are advancing.
Look for whether AP01, for progressive pulmonary fibrosis, and AP02, for idiopathic pulmonary fibrosis, advance. Progress would make one possibility more relevant: if future treatment of the fibrosis also eases associated cough, fewer patients may need Haduvio; delays or setbacks would make that potential effect less immediate. Program progress alone does not show that cough improves, so the key question is whether treatment changes cough in these patients.
Trevi Therapeutics, Inc. earningsThis company
Trevi Therapeutics has no product sales, so this report will show whether its cash can keep funding Haduvio’s clinical development.
Read cash, cash equivalents and marketable securities at quarter-end alongside cash used in operations and research and development spending. Slower cash use with research still advancing would leave more room to fund the studies Trevi has planned; faster use would bring outside financing sooner, with possible dilution or cuts to development. Trevi has said its current funds may support specified planned studies into 2028, but will not fund Haduvio through approval for any indication.
