Company

Company overview

ALX Oncology Holdings Inc. focuses on evorpacept and ALX2004, two cancer drug candidates in clinical development. The business depends heavily on evorpacept, with most development efforts and spending directed toward these two candidates. Neither has completed a pivotal trial, and neither may be marketed before regulatory approval.

The company funds its work through stock offerings and borrowing rather than customer payments. It needs additional capital to advance development. Any future drug-sales revenue would depend on successful trials, regulatory approval, physician prescribing and coverage and reimbursement from insurers and other payors.

ALX Oncology relies on outside organizations to run clinical trials and on contract manufacturers to supply its drugs; it has no manufacturing facilities of its own. Third-party European patents could also limit development of evorpacept for certain cancer uses in some EU territories unless the company obtains a license or the patent barriers are otherwise removed.

  • Competition
  • Shared drivers
  • Supplier
  • Customer
  • Pfizer’s cancer programs inherited from Trillium overlap with ALX Oncology’s evorpacept, while Pfizer also holds an exclusive license to related U.S. SIRPα patent rights. The drugmaker therefore matters both as a rival and through those patent rights, although ALX believes it does not infringe the issued U.S. patent.

  • Bristol Myers Squibb competes with both sides of ALX Oncology’s pipeline: CD47-pathway treatments overlap with evorpacept, and EGFR-targeted antibody-drug conjugates (ADCs) compete with its ADC program. The company already sells medicines for serious diseases; its development updates in those two areas are the news most relevant to ALX, which has no approved products or drug-sales revenue.

  • BioNTech’s EGFR-targeted antibody-drug conjugates (ADCs) compete with ALX Oncology’s emerging ADC pipeline. Although BioNTech’s commercial product is the Comirnaty COVID-19 vaccine, its EGFR-ADC development news is what matters for assessing competition facing ALX’s preclinical ALX2004 program.

  • AstraZeneca’s EGFR-targeted antibody-drug conjugates (ADCs) compete with ALX Oncology’s own EGFR-ADC development approach. AstraZeneca already earns revenue from selling cancer medicines, but its EGFR-ADC research progress is the relevant comparison for ALX’s still-preclinical ALX2004 program.

  • ALX Oncology depends on Eli Lilly to supply ramucirumab for certain evorpacept combination trials; Lilly manufactures and sells medicines. Watch whether that supply agreement continues through trial completion: if it ends early, ALX may need another source of ramucirumab.

  • ALX Oncology uses patent rights assigned by OmniAb’s Crystal Bioscience subsidiary and may owe payments as products reach specified clinical and regulatory milestones. OmniAb earns money by providing drug-discovery technology to partners; ALX’s agreement provides for up to $11.1 million in aggregate milestone payments, with each milestone payable only once across all products.
