Company overview

Tvardi Therapeutics is a clinical-stage drug developer focused on inflammatory diseases and diseases involving abnormal cell growth, including cancer. Its candidates target STAT3, a protein involved in these diseases. The business centers on TTI-101, being tested for hepatocellular carcinoma, the main form of liver cancer, and TTI-109, a related candidate designed to improve drug delivery and tolerability. TTI-109 is in an early trial in healthy volunteers. A TTI-101 trial in idiopathic pulmonary fibrosis did not meet its goals.

No product is approved for sale, and the company has generated no revenue. It has funded development through investor capital and its merger with Cara, and needs additional funding to continue its planned work. Future revenue could come from selling approved drugs or payments from biopharmaceutical partners under licensing or collaboration agreements. Drug sales require successful further trials and regulatory approval, and would also depend on insurance coverage and reimbursement.

Tvardi retains exclusive worldwide rights to its candidates, partly through licenses from Baylor College of Medicine that carry development obligations and royalties. TTI-101 lacks patents protecting the compound itself; its patent protection instead covers particular uses, manufacturing and formulations.

Tvardi's prospects hinge on TTI-101 results in liver cancer (HCC), including combinations with Roche's atezolizumab and bevacizumab, whose established regimen also sets a competitive benchmark.

  • Purple Biotech's cancer candidate NT219 inhibits STAT3, competing with Tvardi's approach to developing cancer drugs around the same target. Further NT219 development depends on a partner or sufficient investment, making funding or partnership news the key sign that this rival can advance.

  • Tyra Biosciences is developing TYRA-430, an oral small-molecule drug for FGF19-positive liver cancer (HCC), overlapping with patients Tvardi targets with TTI-101. Progress in TYRA-430 would show how competition is developing in that specific patient group.

  • Moleculin Biotech's WP1066 portfolio competes with Tvardi in STAT3-directed drug development, rather than directly matching its liver-cancer focus. Moleculin's disclosed clinical work centers on Annamycin, so WP1066-specific progress is more relevant to this tie than its main trial news.

  • Merck's pembrolizumab has a dual role for Tvardi: it competes as a liver-cancer (HCC) treatment and is also combined with TTI-101 in one arm of Tvardi's liver-cancer trial.

  • AstraZeneca's established durvalumab-plus-tremelimumab treatment for liver cancer (HCC) sets a competitive benchmark for Tvardi's TTI-101 development.

  • Bristol Myers Squibb's nivolumab-plus-ipilimumab treatment competes for liver-cancer (HCC) patients in the market Tvardi aims to enter with TTI-101.
