Tenaya Therapeutics is testing treatments aimed at the underlying causes of heart disease. Its priority programs are TN-201, a gene therapy for MYBPC3-associated hypertrophic cardiomyopathy, and TN-401, a gene therapy for PKP2-associated arrhythmogenic right ventricular cardiomyopathy. Both are in early clinical trials.
Tenaya generated no revenue through the end of 2025 and has funded operations mainly by issuing shares. It subsequently entered a research collaboration with Alnylam to identify and validate cardiovascular gene targets, with Alnylam reimbursing research efforts. After validation, Alnylam controls development and commercialization of products aimed at those targets; Tenaya cannot independently research or develop treatments directed at them while they remain collaboration targets.
For its own pipeline, Tenaya plans to bring at least some rare-disease treatments to market itself and seek licensing or development partners for treatments aimed at more common conditions. Product sales depend on completing development and obtaining regulatory approval, and on insurers and other payors covering treatment. Continuing development also requires substantial additional funding.
The risk section is the longest, dullest part of the filing. This keeps only what can really hurt you: where it hits, how hard, when it plays out.
Every company claims an edge. Each one is checked against hard numbers like margins and returns: which hold, and which line gives way first.
Lexeo Therapeutics’ LX2020 competes with Tenaya’s TN-401 for the same patients with PKP2-associated cardiomyopathy; both are developing gene therapies rather than approved treatments. LX2020’s clinical results are therefore a direct comparison point for TN-401.
Rocket Pharmaceuticals’ RP-A601 directly challenges Tenaya’s TN-401 as a gene therapy for PKP2-associated arrhythmogenic cardiomyopathy. Watch RP-A601’s clinical progress: it is another attempt to deliver a working PKP2 gene to the same patient group.
Alnylam Pharmaceuticals pays Tenaya to identify and validate cardiovascular gene targets, then licenses those targets for drug development under their March 2026 collaboration. Advancement of Tenaya-nominated targets matters for potential development, regulatory and sales milestone payments; these are not revenue reported for 2025.
Edgewise Therapeutics’ EDG-7500 overlaps the hypertrophic cardiomyopathy patients Tenaya seeks to treat with TN-201, but uses a different approach: it is designed to change heart-muscle contraction and relaxation, while TN-201 targets the MYBPC3 genetic cause. EDG-7500’s results in those patients help gauge the alternatives TN-201 would face.
Cytokinetics already sells MYQORZO for hypertrophic cardiomyopathy, in a treatment market that overlaps Tenaya’s prospective TN-201 gene therapy. Its MYQORZO sales through U.S. specialist centers and cardiologists offer a concrete view of the market Tenaya hopes to enter.
Braveheart Bio’s BHB-1893 seeks an overlapping hypertrophic cardiomyopathy market with Tenaya’s TN-201, using cardiac myosin inhibition rather than gene therapy. BHB-1893 is Braveheart’s sole candidate, and its development outside Greater China depends on the exclusive license from Hengrui.
Solid Biosciences’ work on gene therapies for inherited heart disease makes its cardiac delivery and safety results relevant to Tenaya’s TN-201 and TN-401. Its SGT-601 program for TNNT2-mediated dilated cardiomyopathy is one concrete program to watch, though it targets a different genetic disease.
Bristol Myers Squibb already sells Camzyos to hypertrophic cardiomyopathy patients who overlap Tenaya’s prospective TN-201 market. Camzyos is an existing treatment alternative to the MYBPC3-targeting gene therapy Tenaya is developing.
Lexicon Pharmaceuticals’ sotagliflozin is a potential competitor to Tenaya’s TN-201 in hypertrophic cardiomyopathy, not an established direct rival to its gene therapy. Sotagliflozin’s progress in that disease is the relevant news for Tenaya, rather than Lexicon’s broader revenue from drug sales, collaborations and licenses.
Alnylam Pharmaceuticals, Inc. earningsALNY
If Alnylam’s report discloses progress on collaboration targets, it will show whether Tenaya’s potential milestone payments are moving closer.
Look for any disclosed progress on cardiovascular targets nominated by Tenaya, and for milestone information tied to that progress. Advancement could bring Tenaya closer to potential development, regulatory or sales milestone payments; without disclosed progress, those potential payments should not be treated as revenue already earned.
Bristol-Myers Squibb Co. earningsBMY
Bristol Myers Squibb’s Camzyos sales will show how patients and providers are taking up an existing hypertrophic cardiomyopathy treatment that would compete for some of the patients Tenaya hopes to treat.
Read Camzyos sales as a signal about the market for an existing hypertrophic cardiomyopathy treatment, not as direct demand data for Tenaya’s candidate. Strong sales would show that an established option is reaching these overlapping patients, giving Tenaya a meaningful alternative to contend with; weak sales would point to limited uptake of this treatment, not prove that patients would choose gene therapy instead.
Solid Biosciences Inc. earningsSLDB
If Solid Biosciences reports clinical progress or safety information for SGT-601, its report will give Tenaya another view of the challenges of delivering gene therapy to the heart.
Look for clinical progress on SGT-601 and any reported safety or heart-delivery information. Encouraging results would provide another positive reference point for delivering gene therapy to the heart; problems would flag development challenges that Tenaya’s TN-201 and TN-401 also need to address. SGT-601 targets a different inherited heart disease, so its results cannot establish how Tenaya’s candidates will perform.
Edgewise Therapeutics, Inc. earningsEWTX
Edgewise Therapeutics’ report will show whether EDG-7500, another approach for hypertrophic cardiomyopathy, is advancing and could become an alternative to Tenaya’s TN-201.
Look for clinical progress or results for EDG-7500 in hypertrophic cardiomyopathy patients. Progress or encouraging results would show that another way to change heart-muscle contraction and relaxation is advancing, giving future TN-201 patients another potential treatment option; setbacks would weaken that alternative, but would not prove TN-201 works.
Rocket Pharmaceuticals, Inc. earningsRCKT
If Rocket Pharmaceuticals reports clinical progress for RP-A601, Tenaya will see how another gene therapy for the same PKP2-associated cardiomyopathy patients is advancing.
Compare RP-A601 with TN-401: both are gene therapies being developed for patients with PKP2-associated cardiomyopathy. If Rocket reports clinical progress, a direct competitor is advancing in the same patient group; if the program hits a setback, that competitive pressure eases, but the news does not establish whether TN-401 is safe or effective.
Tenaya Therapeutics, Inc. earningsThis company
Tenaya Therapeutics has no approved products or product revenue, so this report will show whether its cash use and research spending can support continued development of its heart-disease therapies.
Read cash, cash equivalents and investments alongside cash used in operations and research and development spending. If cash use stays controlled while research continues, Tenaya has more room to develop TN-201 and TN-401; if cash falls as operating use rises, it may need financing sooner and could delay, reduce or stop programs. Tenaya does not break research spending out by candidate, so the total cannot show which therapy drove a change.
