Karyopharm Therapeutics’ business centers on selinexor, sold as XPOVIO in the United States and NEXPOVIO in Europe and the United Kingdom. The drug blocks XPO1, a protein that moves substances out of the cell nucleus, and is used to treat cancers including multiple myeloma. U.S. sales supply most of the company’s revenue. Specialty pharmacies sell the drug to patients, while specialty distributors supply healthcare providers; insurance coverage, reimbursement and patients’ ability to afford copayments affect those sales.
Outside the United States, partners commercialize selinexor in their territories and pay Karyopharm royalties and payments tied to development or commercial milestones. Menarini supplies most of its license and other revenue, with Antengene another partner. Menarini’s arrangement to reimburse development expenses has expired, removing that source of income.
The business depends heavily on XPOVIO’s adoption and on obtaining approvals for additional uses. Karyopharm needs additional capital to complete ongoing selinexor trials. Its U.S. approval for diffuse large B-cell lymphoma requires a confirmatory trial; a missed study milestone puts that approval at risk of withdrawal. The company does not actively promote XPOVIO for that disease, which contributes little revenue.
The risk section is the longest, dullest part of the filing. This keeps only what can really hurt you: where it hits, how hard, when it plays out.
Every company claims an edge. Each one is checked against hard numbers like margins and returns: which hold, and which line gives way first.
Karyopharm depends on U.S. XPOVIO sales, clinical development and access to funding; watch Menarini, an important overseas commercialization partner whose annual development reimbursements ended after 2025.

Karyopharm’s next report will show whether U.S. XPOVIO sales are growing enough to help fund its ongoing drug development.
Read U.S. XPOVIO net sales alongside cash used in operations. Rising sales with lower operating cash use would show the product business is providing more support for development; flat or falling sales with heavier cash use would show revenue is not closing the funding gap and development remains more dependent on outside capital.
