Company

Company overview

Annexon is a clinical-stage drug developer focused on diseases in which the complement immune system contributes to inflammation and damage. Its approach targets C1q, a protein near the start of one complement pathway, with the aim of blocking that pathway while preserving the immune functions of the other two. Its main development programs are tanruprubart for Guillain-Barré syndrome and vonaprument for geographic atrophy, an eye condition.

No Annexon product is approved for commercial sale, and it has no product-sales revenue. Investors, primarily through equity financing, currently fund its operations. Annexon holds worldwide development and commercialization rights to its candidates. It plans to sell approved medicines itself in markets it can serve with a focused sales team and explore partnerships elsewhere. Future sales would depend in part on government and private insurers covering treatment and providing adequate reimbursement.

The business depends on completing development and securing regulatory approval before medicines can be sold. For tanruprubart, the FDA has indicated that the evidence supporting the applicability of overseas trial results to Western patients may require additional patient data. Annexon also relies entirely on outside manufacturers to produce its candidates.

Annexon’s value hinges on clinical and regulatory success in Guillain-Barré syndrome and geographic atrophy, with Astellas and Regeneron also competing in geographic atrophy.

  • ADARx Pharmaceuticals identifies Annexon as a competitor in geographic atrophy, making it worth watching alongside Annexon’s late-stage vonaprument program for that eye condition. ADARx is developing agazisiran for diseases driven by the immune system’s complement activity; progress on that candidate is the relevant development to follow.
